Astaxanthin production by Phaffia rhodozyma using a pH-stat control
Fed-batch essays using yeast Phaffia rhodozyma were performed to obtain high astaxanthin concentrations. The yeast was grown over an inexpensive media obtained from the Yucca fillifera date. Feeding was carried out through implantation of a pH-stat control system. Biomass (28.7 mg/l) and pigment (18 mg/l) concentration three folded those that were obtained by previous batch cultures.
متن کاملBiogeography, host specificity, and molecular phylogeny of the basidiomycetous yeast Phaffia rhodozyma and its sexual form, Xanthophyllomyces dendrorhous.
Phaffia rhodozyma (sexual form, Xanthophyllomyces dendrorhous) is a basidiomycetous yeast that has been found in tree exudates in the Northern Hemisphere at high altitudes and latitudes. This yeast produces astaxanthin, a carotenoid pigment with biotechnological importance because it is used in aquaculture for fish pigmentation. We isolated X. dendrorhous from the Southern Hemisphere (Patagonia...
متن کاملStructural and phylogenetic analysis of the actin gene from the yeast Phaffia rhodozyma.
The gene coding for actin from Phaffia rhodozyma was cloned and sequenced. The Phaffia actin gene contains four intervening sequences and the predicted protein consists of 375 amino acids. The structural features of the Phaffia actin introns were studied and compared with actin introns from seven fungi and yeasts with ascomycetous and basidiomycetous affinity. It was shown that the architecture...
متن کاملSelection of Astaxanthin-Overproducing Mutants of Phaffia rhodozyma with beta-Ionone.
beta-Ionone, an end ring analog of beta-carotene, inhibits astaxanthin production in the red yeast Phaffia rhodozyma. Astaxanthin-overproducing mutants of this yeast are easily spotted on beta-ionone-containing yeast malt agar plates. beta-Ionone appears to block astaxanthin synthesis at the beta-carotene level.
متن کاملOverexpression of a bifunctional enzyme, CrtS, enhances astaxanthin synthesis through two pathways in Phaffia rhodozyma
BACKGROUND A moderate-temperature, astaxanthin-overproducing mutant strain (termed MK19) of Phaffia rhodozyma was generated in our laboratory. The intracellular astaxanthin content of MK19 was 17-fold higher than that of wild-type. The TLC profile of MK19 showed a band for an unknown carotenoid pigment between those of β-carotene and astaxanthin. In the present study, we attempted to identify t...
